Applications of BBa_K3682001

We used pepP to construct two kinds of protein. One is pepP sequence with a TEV cleavage site and 6x histag behind it. The other one(part BBa_K3682008) is pepP sequence with hACE2 sequence(part BBa_K3682002) and also having a TEV cleavage site and 6X histag. We used our bacterial expression vector(part BBa_K3682000) to enclose and express pepP protease. We used gibson assembly to assemble the parts and the got positive results(Figure 2).

After plasmid construction, we transformed the plasmid into dH5alpha and BL21 strain to replicate the plasmid and express the protein. We use SDS page and western blot to check the results and the size of the protein is right.
